Resource or Project Abstract
Large quantities of styrene are used annually in the petro-chemical and polymer processing industries leading to the release of styrene into the environment and the generation of significant quantities of styrene contaminated waste. This poses a significant threat to the environment and the associated potential toxic effects on human health, including carcinogenesis, are a cause of concern. However, a number of microbial species have the ability to degrade styrene, particularly members of the bacterial genus, Pseudomonas, and this has led to significant interest in using microbial degradation to provide a potential solution to styrene waste remediation. The aim of the current project is to improve the rate of flux through the styrene degradation pathway in Pseudomonas putida CA-3 through recombinant DNA manipulations, thus potentially leading to increased styrene degradation.

A number of styrene degrading Pseudomonas strains, including Pseudomonas putida CA-3, have been well-characterised at both the genetic and physiological level, and a significant body of work has identified the catabolic genes involved in the transformation of styrene to phenylacetic acid, and the subsequent transformation of phenylacetic acid to generate cellular energy, as well as aspects of the genetic regulation and physiological control of the process. Recently, it has also been reported that our P. putida CA-3 strain has the ability to redirect styrene catabolism to the production of a biodegradable polymer, known as poly-hydroxyalkanoate (PHA), under appropriate growth conditions, thus promoting the idea of generating a value-added product from a toxic waste material. This project aims to exploit the considerable knowledge base already generated about bacterial styrene degradation and apply recombinant DNA strategies to potentially improve the styrene degradtion capacity and further charaterise the degradation pathway in P. putida CA-3. |
